Mass Effect is probably one of the best RPG's released in the last years for both console and PC. The PC version has the DLC for free, but it's protected by SecuROM online activation. The XBox 360 version will require extra payment for the DLC. Gears of War series has a great MP, excellent story in SP, but it can be damn frustrating in SP campaign while you will have to constantly save your dumb AI companions. Prince of Persia 2008 deserves the money (including the DLC). GTA 4, I'd wait for the second announced DLC pack, as Rockstar announced to put the both DLC on a disc for sale. The DLC's add quite a bit of extra hours of gameplay, so it will deserve the wait. GTA 4 it's better than San Andreas from so many points orf view (story, voice acting, music, gameplay).
Other recommendations: Dead Space, Mirror's Edge, Assassin Creed (control wise it's better than its PC counterpart).
Bullet Witch it's a bit of controversial game: some loved it, some hate it, so check trailers and reviews plenty to see if you might like it. COD 4: Modern Warfare had an excellent port, so you might want to check it. Condemned series might worth a look.
Dark Messiah Might and Magic: Elements and Rise of the Argonauts are decent, along with Spectral Force 3: Innocent Rage, Tales of Vesperia, Kingdom Under Fire: Circle of Doom, The Last Remnant, Eternal Sonata, Far East of Eden Ziria: Tales from Distant Jipang, Enchanted Arms, Diario: Rebirth Moon Legend and Star Ocean: Last Hope (which are some great JRPGs, if you like the genre).
Star Wars: The Force Unleashed falls in the category of either you hate it, either you love it, so again my recommnedation it's to check trailers, reviews, gameplay movies on youtube (the site it's a good source for seeing the in-game action).
If you are a fan of the Koei games, you might want to grab the ones that you don't have on your PS2 (Dynasty Warriors 6 and Dynasty Warriors 6: empires, plus Warriors Orochii 2). Marvel Ultimate Alliance Gold it's better than any version out there (10 extra characters are great in my book) and MUA2 it's going to be launched soon. Another game that will be launched soon it's Red Faction: Guerrilla. Also coming soon are Venetica andDivinity2 (RPG's, one developped by the same people which created Drakensang: the Dark Eye on PC, the other from the same people that gave us Divine Divinity and Beyond Divinity on PC). Sacred2 it seems to be better polished on XBox 360. Fallout 3 and Oblivion are present on XBox 360, but I'd rather have a heavily modded PC version, than the plain XBox 360 or PS 3 version (though the y do have the official DLC).
Alpha Protocol and Brave: A Warrior's Tale should be on the buying list ( though the games will be launched this autumn). Also Wet should come out this autumn.
XBox 360 has an excellent catalogue of racing games, if you like those: Test Drive Unlimited (6 car DLC's, exclusive for XBox 360), Juiced2: Hot Import Nights, Fuel, Ridge Racer 6, Forza Motorsport 2+3, FlatOut: ultimate Carnage, Project Gotham Racing 3+4, Dirt and Dirt 2 (the second will be launched in september), NFS from Carbon to Undercover,Midnight Club: Los Angeles, Burnout Revenge, Burnout Paradise GRID and Race PRO.
Fighting games on XBox 360 comprise: Tekken 6, DOA4, Soul Calibur 4, Street Fighter 4, Mortal Kombat vs DC Universe, Dead or Alive 2: Xtreme Edition, DOA4 and King of Fighters 12.
Saints Row series on XBox 360 are a lot better than the PC versions, so if you like GTA style game, that might worth to check them out. Red Dead Redemption from Rockstar should be on your buy list if you like the Wild West as a setting for a shooter (the game should come out this year and it boast an open world, just like in GTA).
Ninety-Nine Nights 1 and the incoming sequel worth to be checked. Onechanbarra provides a bit of mindless fun (though the reviews were abyssmal, so check youtube, the reviews and the trailers). Devil may Cry 4, Ninja Gaiden 2 and Tenchu Z are another action adventures highly recommended.
The Orange Box (this is a must for its Team Fortress 2), Wheelman, X-Blades, TimeShift, Prototype, Damnation, Battlefield: Bad Company, Army of Two, FEAR 1 and 2, Lost Planet: Colonies, Resident Evil 5 , Spiderman: Web of Shadows, Spiderman: Friend or Foe, Unreal Tournament 3, Left for Dead, FarCry 2, The Chronicles of Riddick: Assault on Dark Athena, Call of Juarez series, Call of Duty (you have 2, 3 and World at War plus the already mentioned Modern Warfare), Brothers in Arms: Hell's Highway, Alone in the Dark 2008, Tom Clancy's GRAW 1 + 2, Tom Clancy's Rainbow Six: Vegas 1 + 2, Splinter Cell: Double Agent, Tom Clancy's Ghost Recon 4 are some good choices for Action and FPS.
Strategy games on XBox 360: Sid Meier's Civilization Revolution, Warhammer: Battle March, C&C3 plus its Kane's Wrath expansion, C&C: RA3 , Universe at War: Earth Assault, Tom Clancy's EndWar, Overlord 2
Some nice arcade air simulators: Ace Combat 6 - Fires of Liberation, Blazing Angels series, Tom Clancy's HAWX
